| Description | This enzyme immunoassay is a sandwich assay fort he quantitative determination of anti-epidermal transglutaminase antibodies (IgA anti-heTG). The wells of the microtiter plate are coated with the antigen. In a first incubation step, the anti-heTG antibodies are bound to the coated antigen. To remove all unbound substances, a washing step is carried out. In a further incubation step, a peroxidase-labeled antibody is added. After another washing step to remove all unbound substances, the solid phase is incubated with peroxidase substrate, tetramethylbenzidine (TMB). Finally, an acidic stop solution is added to terminate the enzymatic reaction, whereby the colour changes from blue to yellow. The entensity of the yellow colour is directly proportional to the anti-epidermal-transglutaminase antibody concentration. A dose response curve of the absorbance unit (optical density, OD at 450 nm) vs. concentration is generated, using the values obtained from the standards. IgA anti-epidermal-transglutaminase antibodies present in the samples are determinate directly from this curve. |
